Résumé (français)  :

OBJECTIFS : étudier si l’utilisation d’ondansétron pendant la grossesse est associée à une augmentation du taux de malformations majeures ou usb-groupées.

METHODES :
Les bases de données PubMed / MEDLINE, Cochrane et Reprotox® ont été interrogées. Des études observationnelles comprenant un groupe exposé et un groupe témoin (en bonne santé et / ou correspondant à une maladie) ont été incluses.

RÉSULTATS :
Aucune augmentation significative du risque de malformations majeures, de malformations cardiaques, de fentes orofaciales, de malformations génito-urinaires ou d’hypospadias n’a été identifiée dans notre analyse principale. Une hétérogénéité significative existait pour les fentes palatines isolées. Des estimations ponctuelles élevées et une signification statistique altérée étaient présentes pour certains des résultats parmi les analyses secondaires.

CONCLUSIONS :
L’utilisation de l’ondansétron pendant la grossesse n’a pas été associée à une augmentation significative du taux de malformations majeures ou sélectionnées de malformations dans notre analyse principale. Cependant, les résultats des analyses secondaires justifient la nécessité d’une surveillance continue. Ces résultats peuvent être rassurants pour les femmes enceintes chez qui l’utilisation de l’ondansétron est cliniquement indiquée, car le risque absolu de problèmes possibles semble être faible.

Abstract (English)  :

AIMS: To investigate whether ondansetron use during pregnancy is associated with increased rates of major or subgroups of malformations.
METHODS:
PubMed/MEDLINE, Cochrane and Reprotox® databases were searched. Observational studies comprising an exposed and control group (healthy and/or disease-matched) were included.
RESULTS:
No significant increased risk for major malformations, heart defects, orofacial clefts, genitourinary malformations or hypospadias were identified in our primary analysis. A significant heterogeneity existed for isolated cleft palate. Elevated point estimates and altered statistical significances were present for some of the outcomes among secondary analyses.
CONCLUSIONS:
Ondansetron use during pregnancy was not associated with a significant increase in rate of major or selected subgroups of malformations in our primary analysis. However, results of the secondary analyses warrant the need for continued surveillance. These results may be reassuring for pregnant women in whom ondansetron use is clinically indicated since the absolute risks of possible concerns appear to be low.

Objetivos: Investigar se o uso do ondansetron durante a gravidez está associado a taxas aumentadas de grandes ou subgrupos de malformações.

MÉTODOS:
Foram pesquisadas as bases de dados PubMed / MEDLINE, Cochrane e Reprotox®. Estudos observacionais que incluíram um grupo exposto e controle (saudáveis ​​e / ou pareados por doenças) foram incluídos.

RESULTADOS:
Não foi identificado risco aumentado significativo para malformações importantes, defeitos cardíacos, fendas orofaciais, malformações geniturinárias ou hipospádias em nossa análise primária. Existia uma heterogeneidade significativa para fenda palatina isolada. Estimativas pontuais elevadas e significâncias estatísticas alteradas estavam presentes em alguns dos resultados entre as análises secundárias.

CONCLUSÕES:
O uso de ondansetrona durante a gravidez não foi associado a um aumento significativo na taxa de subgrupos principais ou selecionados de malformações em nossa análise primária. No entanto, os resultados das análises secundárias justificam a necessidade de vigilância continuada. Esses resultados podem ser tranquilizadores para mulheres grávidas nas quais o uso do ondansetron é clinicamente indicado, pois os riscos absolutos de possíveis preocupações parecem ser baixos.
